Mice were xenografted with the human lung adenocarcinoma line A549 or large cell lung cancer line FT821. Tumour uptake of 18F-FLT and 18F-FDG was imaged using PET/CT before and 1 week after irradiation. In A549 tumours, 18F-FLT uptake was significantly decreased, and 18F-FDG uptake was unchanged post-irradiation compared with pre-irradiation. In FT821 tumours, uptake of both 18F-FLT and 18F-FDG uptake was substantially decreased post-irradiation compared with pre-irradiation. In both xenografts, tumour volumes in the irradiated groups were significantly decreased compared with those in the control group. 18F-FLT is expected to contribute to individual NSCLC therapy because it accurately evaluates the decrease in tumour activity that cannot be captured by 18F-FDG. 18F-FDG may be useful for evaluating surviving cells without being affected by the inflammatory reaction at an extremely early stage, approximately 1 week after irradiation. Combined use of 18F-FLT and 18F-FDG PET/CT imaging may increase the accurate prediction of radiotherapy efficacy, which may lead to improved patient outcomes and minimally invasive personalised therapy. In the present study, the proliferation of recA-deficient Escherichia coli strains was revealed to be inhibited by 1% L-histidine under aerobic conditions. This inhibition of proliferation was not observed under anaerobic conditions, indicating that L-histidine enhances oxidative DNA damage to E. coli cells. Reverse transcription-quantitative polymerase chain reaction analysis demonstrated that the expression of recA in E. coli MG1655 increased ~7-fold following treatment with 10 mM hydrogen peroxide (H2O2) plus 1% L-histidine, compared with that following exposure to H2O2 alone. L-histidine increased the genomic fragmentation of E. coli MG1655 following exposure to H2O2. In addition, L-histidine increased the generation of intracellular hydroxyl radicals in the presence of H2O2 in E. coli cells. Next, our group investigated the disinfection properties of the H2O2 and L-histidine combination. The combination of 100 mM H2O2 and 1.0% L-histidine significantly reduced the number of viable cells of extended-spectrum-β-lactamase-producing E. coli and multidrug-resistant Pseudomonas aeruginosa, and this treatment was more effective than 100 mM H2O2 alone, but this effect was not observed in methicillin-resistant Staphylococcus aureus or vancomycin-resistant Enterococcus faecium. The combination of L-histidine and H2O2 may be a useful strategy to selectively increase the microbicidal activity of oxidative agents against Gram-negative bacteria.","ja":"Excessive damage to DNA and lipid membranes by reactive oxygen species reduces the viability of bacteria. We created such a model, and we performed PET/CT. The mice were divided into two groups according to tumor volume: a small-tumor group (<20 mm(3)) and a large-tumor group (>20 mm(3)). We reconstructed the following conditions based on list-mode data: non-gated (3D) images and gated (4D) images, divided based on the respiratory cycle (expiration phase, stable phase, and inspiration phase). We calculated the maximum standardized uptake values (SUVmax) in each phase. We used the % difference [= (4D SUVmax - 3D SUVmax)/3D PET SUVmax × 100 (%)] to evaluate the differences in the 4D SUVmax and 3D SUVmax. The 4D SUVmax values were significantly higher than the 3D SUVmax, regardless of the tumor size. The % difference for the small tumors was greater than that for the large tumors, and it was highest in the stable phase. We conclude that the SUVmax in the stable phase under respiratory-gated PET are the most reliable. The SUVmax observed under non-gated PET are considered to be more frequently underestimated in cases involving small tumors than in those involving large tumors. In the chronologic study evaluating the time course of tumor development, the size of the tumor is small in early stage, and respiratory-gated PET is effective in reducing the underestimation of such tumors caused by respiratory motion.","ja":"Our aim in this study was to clarify the effects of respiratory-gated PET in the evaluation of lung cancer according to the (18)F-FDG uptake in an orthotopic transplantation mouse model. Herein we describe the development of multifunctional core-shell Fe(3)O(4)@SiO2 nanoparticles with the ability to target inflammatory endothelial cells via VCAM-1, magnetism, and fluorescence imaging, with efficient magnetic resonance imaging contrast characteristics. Superparamagnetic iron oxide and fluorescein isothiocyanate (FITC) were loaded successfully inside the nanoparticle core and the silica shell, respectively, creating VCAM-1-targeted Fe(3)O(4)@SiO2(FITC) nanoparticles that were characterized by scanning electron microscopy, transmission electron microscopy, fluorescence spectrometry, zeta potential assay, and fluorescence microscopy. The VCAM-1-targeted Fe(3)O(4)@SiO2(FITC) nanoparticles typically had a diameter of 355 ± 37 nm, showed superparamagnetic behavior at room temperature, and cumulative and targeted adhesion to an inflammatory subline of human umbilical vein endothelial cells (HUVEC-CS) activated by lipopolysaccharide. Further, our data show that adhesion of VCAM-1-targeted Fe(3)O(4)@SiO2(FITC) nanoparticles to inflammatory HUVEC-CS depended on both shear stress and duration of exposure to stress. Analysis of internalization into HUVEC-CS showed that the efficiency of delivery of VCAM-1-targeted Fe(3)O(4)@SiO2(FITC) nanoparticles was also significantly greater than that of nontargeted Fe(3)O(4)@SiO2(FITC)-NH2 nanoparticles. Removal rates of 131I of approximately 76 ± 3% after further washing with 1.2 wt% Na2S2O4 for 2 days and 47 ± 2% washing with 10 wt% Na2S2O4 for 2 days were obtained. These values agreed with the dissolution rates of 131I in the liquid used for washing within 2 to 3% error. The removal rates of 134Cs and 137Cs after washing with 1.2 wt% Na2S2O4 for 2 days were 69 ± 47% and 100 ± 52%, respectively, which were in poor agreement with the corresponding dissolution rates. The removal rate of 131I after brief washing with 10 wt% Na2S2O4 was negligible and the dissolution rate was 5 ± 2%. In this study, fluorescent silica NPs were synthesized by a modified Stöber method using conjugates of 3-mercaptopropyltrimethoxysilane (MPS) and maleimide-fluorescein isothiocyanate (maleimide-FITC). Mean diameters of the NPs were controlled between 212-2111 nm by regulating MPS concentration in the reaction mixture. Maleimide-FITC molecules were doped into NPs or conjugated to the surface of NPs through the chemical reaction of maleimide and thiol groups. The data showed that the former NPs are better than the latter by comparing their fluorescence intensity. Furthermore, folate molecules were linked to the FITC-doped silica NPs by using polyethylene glycol (PEG) (NH2-PEG-maleimide) as a spacer, thus forming folate receptor targeting fluorescent NPs, referred to as NPs(FITC)-PEG-Folate. The quantitative analysis of cellular internalization into different cancer cells showed that the delivery efficiency of KB cells (folate receptor-positive cells) is more than six-fold higher than that of A549 cells (folate receptor-negative cells). The delivery efficiency of KB cells decreased significantly after free folate addition to the cell culture medium because the folate receptors were occupied by the free folate. The NPs endocytosis mechanism was also investigated. It was shown that clathrin, an inhibitor of cell phagocytosis, markedly decreased the NPs uptake into KB cells, suggesting that it plays an important role in NPs cellular internalization. These results demonstrated that the novel particles of NPs(FITC)-PEG-Folate are promising for fluorescent imaging or targeting delivery to folate receptor-positive tumors.","ja":"Multifunctionalized nanoparticles (NPs) are emerging as ideal tools for gene/drug delivery, bioimaging, labeling, or intracellular tracking in biomedical applications, and have attracted considerable attention owing to their unique advantages. The amount of H(2) produced depended on the amount of Ag NPs on the clay and the concentration of 2-propanol in the suspension. The photogenerated holes indicate a reaction with OH(-) because the generation of ()OH radicals decreased in the presence of 2-propanol, as observed using a spin-trapping electron spin resonance (ESR) method. The amount of H(2) produced depended on the amount of Ag NPs on the clay and the concentration of 2-propanol in the suspension. Antibacterial activity of Ag NPs on clay was also observed against Escherichia coli even after 12years. This simple method of preparation in n-hexanol led to the successful formation of highly stable and photoactive Ag NPs on montmorillonite clay."},"publication_date":"2010-04-08","publication_name":{"en":"Journal of Colloid and Interface Science","ja":"Journal of Colloid and Interface Science"},"volume":"345","number":"2","starting_page":"433","ending_page":"441","languages":["eng"],"referee":true,"identifiers":{"doi":["10.1016/j.jcis.2010.01.034"],"issn":["0021-9797"]},"published_paper_type":"scientific_journal"},"priority":"input_data"}

However, since dose limits for contamination by medical examinations have not been set, management of dose measurements and contamination records is called for. With increasing use of the IVR technique, reports of radiation injury and the symptoms associated with it have become more common. To advance our understanding of this situation and to reduce contamination, it is necessary to carry out contamination management. The reflection film on which colors are formed by irradiating X-rays has recently come into use. Dose measurement is possible with the use of this film, and, because effective results can be obtained as a result of performing fundamental examinations, the film actually provides dose measurements for the IVR technique. Another benefit is that maximum patient skin dose and dose distribution can be determined in addition to dose measurement. Moreover, since various methods were examined in this study, the method of dose evaluation is also reported for those wishing to employ it in the clinical setting.","ja":"照射部位が多彩で，X線管-皮膚間距離が不定なIVR手技に対してX線照射により直ちに発色する感材で作られた低感度の反射型フィルムを用いた被曝線量測定について検討した結果，良好な結果が得られたので報告した．"},"publication_date":"2003-01-20","publication_name":{"en":"Japanese Journal of Radiological Technology","ja":"日本放射線技術学会雑誌"},"volume":"59","number":"1","starting_page":"121","ending_page":"129","languages":["jpn"],"referee":true,"identifiers":{"doi":["10.6009/jjrt.kj00000921601"],"issn":["0369-4305"]},"published_paper_type":"scientific_journal"},"priority":"input_data"}

After evaluating two methods for compensating relaxation times, we selected a long TR, short TE sequence to achieve better reproducibility. The cerebral blood flow (CBF) value was measured by scintigraphy following the method of quantification reported in the previous literature. N-acetyl aspartate (NAA) concentrations and CBFs value were decreased statistically (P < 0. 05) compared with those of the age-matched normal controls. However, the decreased rate of NAA concentration was changed more than the CBF value, and no linear correlation was found between the two values. We considered that the NAA concentration was not always correlated with that of the present cerebral perfusion. The NAA concentration showed a more dispersive distribution in patients than in controls, indicating a large individual variation in neuronal impairment. The Cho/Cr (creatine and phosphocreatine) ratio of meningioma was considerably higher than that of glioma of all grades. We have experienced only two cases of ependymoma and the Cho/Cr ratios of both were lower than that of glioma. It seems likely that 1H-MRS can be used to differentiate Grade 2 from Grade 4 in most cases of astrocytoma based on the NAA/Cho ratio, though a few cases will overlap. Meningioma can be distinguished easily from glioma, and the results of our study suggest that ependymoma shows a characteristic pattern on 1H-MRS, different from those of other brain tumors.","ja":"We studied the feasibility of characterizing brain tumor tissue by localized proton magnetic resonance spectroscopy (1H-MRS).
