2.	Makoto Ohashi and Takahito Saito : Practical study for future establishment of sustainable society as the cultural education, Journal of University Education Research, Vol.13, No.13, 56-62, 2016. (Summary) Personnel training for global society is one of the important issues for the innovation of university education. A stand point of view for the establishment of sustainable society is considered to have a potential role for understanding issues of global society. To study this relationship, concepotional understanding for global system is indispensable and it closely related to the object of holistic education. Here, we established a holistic educational program consisted with group study by the conbination of students, citizens and foreign studetns. Practical tranings based on the traditional agriculture and medicine are employed as the optional lecture. In this report, we discussed on the effect of the practical study for the future establishment of sustainable society by employing our educational program. (Summary) Various animal milks and their dairy products like AIRAG (fermented horse milk) and yogurt are very popular among Mongolian people. The climate of Mongolia is very severe, and they preserve their health by the intake of above animal milks and dairy products. In this study, we explored the diversity of bacteria in Mongolian animal milks (Cow, Horse, Goat, and Camel), AIRAG, and Camel milk yogurt by the clone library method of their 16S rRNA genes.13;Firstly, we prepared the whole genomic DNA from four animal milks and two dairy products, and amplified each 16S rRNA genes by PCR. PCR products (about 1.4kbp) were cloned into pGEM-T vector, and analyzed DNA sequences of totally 79 clones from AIRAG, Camel milk yogurt, and four animal milks. It was revealed that homologous clones to Lactobacillus helveticus are dominant in the clone libraries of both dairy products, whereas the other clones to Lactococcus from AIRAG, and Acetobacter from camel milk yogurt. Furthermore, We found that homologous clones to Lactococcus are dominant in Mongolian cow and horse milks, whereas Leuconostoc in camel milk. Therefore, it might be useful information for screening the bioactive strains from milk products in Mongolia. (Summary) 徳島大学全学共通教育では，平成17 年度より地域の社会人を活用して体験を取り入れた共創型授業を行っている．『身近な福祉を見て・知って考えてみる』では，障害者福祉だけでなく，教育改革，生命倫理，太平洋戦争，日本国憲法など，幅広い視野から福祉を考える授業を，社会人を加えた共創型学習科目として実施している．通常の授業では，学生の側から意見を出し合うことには限界があるが，今回の共創型学習科目は，学生の側から出た意見を中心に，社会人を交えて討論する形で展開した．また，『「つたえること」と「ものづくり」―科学で遊ぼう―』，『「つたえること」と「ものづくり」―あいのメッセージ―』では，ものづくりを素材としたグループ学習を行い，必要に応じて社会人を含めた討論を行う形式の授業を行った．その結果，様々な項目において教員とは違った視点からの意見を学生から引き出す役割を社会人が担っていることがわかった．また，このような社会活用の効果を専門的人材の不足する開発途上国の教育においても有効であることを検証する目的で，ソロモン諸島国マライタ島の小学校において，現地の定年退職者などを活用した共創型授業を行った．その結果，児童たちだけではなく，教員や社会人自身にも良い刺激となり，地域社会に貢献できる新たな教育法であることが示された． (Keyword) 地域 / 社会人活用 / 共創型学習 (Tokushima University Institutional Repository) ● Metadata: 82671 (Link to Search Site for Scientific Articles) ● CiNii @ National Institute of Informatics (CRID): 1050845762393910528 (Tokushima University Institutional Repository: 82671, CiNii: 1050845762393910528)

(Summary) Supplementary lessons in Mathematics and Sciences were introduced in the general education of Tokushima University from 2003 in order to seal the gap of fundamental studies between the courses of high school and University. The main purpose of these lessons is to cultivate the novel interests on these subjects for students themselves by using textbooks for high school students. The results of student evaluation survey revealed the potential needs of supplementary lessons in other subjects and reevaluation for crowded curriculum of sciences in the course of high school. A designed new curriculum is indispensable including supplementary lessons in other subjects for a coming society with fewer children in Japan. Establishment of systematic course of general education is needed including the fundamental studies by the teaching staff of the university. Further development of the supplementary lessons should be undertaken for students various in their understanding levels or interests. We conduct ananonymous survey in the form of a questionnaire at the end of class in order to compare the motivation forlearning or understanding between the students of four departments. Junko Nio, Wakako Fujimoto, Akihikro Konno, Yasuhiro Kon, Makoto Ohashi and Toshihiko Iwanaga : Cellular expression of murine Ym1 and Ym2, chitinase family proteins, as revealed by in situ hybridization and immunohistochemistry, Histochemistry and Cell Biology, Vol.121, No.6, 473-482, 2004. (Summary) Ym is one of the chitinase family proteins, which are widely distributed in mammalian bodies and can bind glycosaminoglycans such as heparin/heparan sulfate. Ym1 is a macrophage protein produced in parasitic infections, while its isoform, Ym2, is upregulated in lung under allergic conditions. In the present study, we revealed the distinct cellular expression of Ym1 and Ym2 in normal mice by in situ hybridization and immunohistochemistry. Ym1 was principally expressed in the lung, spleen, and bone marrow, while Ym2 was found in the stomach. Ym1-expressing cells in the lung were alveolar macrophages, and the immunoreactivity for Ym1 was localized in rough endoplasmic reticulum. In the spleen, Ym1-expressing cells gathered in the red pulp and were electron microscopically identified as immature neutrophils. In the bone marrow, immature neutrophils were intensely immunoreactive, but lost this immunoreactivity with maturation. Moreover, needle-shaped crystals in the cytoplasm of macrophages, which formed erythroblastic islands, also showed intense Ym1 immunoreactivity. Ym2 expression was restricted to the stratified squamous epithelium in the junctional region between forestomach and glandular stomach. The function of Ym1 and Ym2 is still unclear; however, the distinct cellular localization under normal conditions suggests their important roles in hematopoiesis, tissue remodeling, or immune responses as an endogenous lectin. (Summary) Superoxide plays a crucial role in innate immunity to various pathogens. We examined the role of superoxides in the transmission of malaria using gp91phox knockout (X-CGD) mice that lack the ability to produce superoxide. Mosquitoes that fed on X-CGD mice infected intraperitoneally with Plasmodium berghei NK65 ANKA formed more oocysts than did those that fed on control mice at any day after infection. The number of oocysts peaked on day 5 post-infection in X-CGD and control mice and then decreased significantly after day 5 post-infection. However, on day 7 post-infection, the infectivity of gametocytes in X-CGD mice was significantly higher than that in control mice. These results show that two pathways, superoxide-dependent and -independent, are involved in the host systems regulating the transmission of malaria and inhibiting gametocyte development. Makoto Ohashi, Masakazu Harada, Setsuo Suguri, Hiroshi Ohmae and Akira Ishii : The role of saliva of Anopheles stephensi in inflammatory response: Identification of a high molecular weight neutrophil chemotactic factor, Parasitology Research, Vol.87, No.5, 376-382, 2001. (Summary) Mosquito bites can elicit dermal hypersensitivity reactions, but little is known about the chemotactic factors for host leukocytes in mosquito saliva. In this study, we determined that saliva from a malarial vector mosquito, Anopheles stephensi, possesses intense neutrophil chemotactic activity. In contrast, the midgut extract had only marginal neutrophil chemotactic activity. Eosinophil chemotactic activity was detected in the midgut but not in the saliva. According to the results of size-exclusion HPLC on a G3000SW column and Western blot analysis, the apparent molecular weight (MW) of the main neutrophil chemotactic factor (NCF) was estimated to be 200 kDa. NCF could bind with IgG from the pooled serum of Solomon islanders, whereas not with that of healthy Japanese. NCF activity was increased upon heating to 56 degrees C for 30 min or protease digestion, whereas it was affected by periodate treatment. Protease-digested NCF and naive NCF bound to lentil lectin-Sepharose, and both were eluted with a competitive sugar, methyl-alpha-D-glucoside. These results indicate that A. stephensi saliva-derived NCF is a high MW glycoprotein, and its protein moiety is important for neutrophil chemotactic activity. This NCF is thought to contribute to the inflammatory reactions through the accumulation of neutrophils at the site of the mosquito bite. Makoto Ohashi, Hiroyuki Arita and Naoko Hayai : Identification of a novel eosinophil chemotactic cytokine (ECF-L) as a chitinase family protein, The Journal of Biological Chemistry, Vol.275, No.2, 1279-1286, 2000. (Summary) A novel eosinophil chemotactic cytokine (ECF-L) was purified from the culture supernatant of splenocytes of mice by a combination of anion-exchange chromatography, Procion red-agarose affinity chromatography, size exclusion high performance liquid chromatography (HPLC), and reverse phase HPLC. The NH(2)-terminal amino acid sequence was determined by direct protein sequencing. An ECF-L cDNA clone of 1,506 nucleotides was isolated from a cDNA library, and the nucleotide sequence predicted a mature protein of 397 amino acids. A recombinant ECF-L showed a level of eosinophil chemotactic activity comparable with that of natural ECF-L, and the activity was inhibited by a monoclonal antibody to ECF-L. ECF-L also attracted T lymphocytes and bone marrow polymorphonuclear leukocytes in vitro, whereas it caused selective extravasation of eosinophils in vivo. ECF-L mRNA was highly expressed in spleen, bone marrow, lung, and heart. A comprehensive GenBank data base search revealed that ECF-L is a chitinase family protein. ECF-L retains those amino acids highly conserved among chitinase family proteins, but Asp and Glu residues essential for the proton donation in hydrolysis were replaced by Asn and Gln, respectively. Although ECF-L contains a consensus CXC sequence near the NH(2) terminus akin to chemokine family proteins, the rest of ECF-L shows poor homology with chemokines. Makoto Ohashi, Y Horii and A Ishii : IgE response in schistosomiasis japonica: characterization of a cercarial allergen in comparison with purified egg allergens., Journal of Helminthology, Vol.71, No.3, 221-226, 1997. (Summary) The relationship between the cercarial allergen and two previously isolated egg allergens (J1, J2) of Schistosoma japonicum was examined especially in terms of the cross-reactivity between them. Semi-purified cercarial allergen (JAC) was obtained from the crude extract of S. japonicum cercariae by gel chromatography on Sephadex G-200. The apparent molecular weight of JAC was estimated approximately as 60-100 kDa. JAC could bind to Con A-Sepharose, indicating its glycoprotein nature. Three groups of BALB/c mice were immunized with JAC , J1 or J2 using A1(OH)3 as adjuvant, and the cross-reactivity of each anti-serum was examined by PCA. Anti-JAC, anti-J1 or anti-J2 serum was highly specific to the corresponding antigen. When IgE-ELISA of S. japonicum patient sera was performed using JAC, J1 or J2 as an antigen, the correlation between anti-J1 and anti-J2 (r = 0.78) was high, whereas the correlation between anti-JAC and anti-J1 (r = 0.27) or between anti-JAC and anti-J2 (r = 0.12) was low. These results suggest that most IgE epitopes on cercarial allergen are independent from those on egg allergens in S. japonicum. (Summary) Infection of mice with Plasmodium berghei engendered a temporary appearance of granulocyte-macrophage colony-stimulating factor (GM-CSF) in the serum. The peak of GM-CSF levels was detected at day 2 post-infection, and then gradually decreased. On the other hand, the number of committed stem cells for granulocytes and macrophages (CFU-GM) in bone marrow transiently decreased at day 2 post-infection, and then increased and peaked at day 6 post-infection. When the serum of P. berghei-infected mice was fractionated by gel chromatography on Sephacryl S-300, GM-CSF activity was detected as a single peak with an apparent molecular weight of 64 KDa. GM-CSF was entirely adsorbed to concanavalin A-Sepharose 4B affinity chromatography, and was sensitive to pronase digestion, indicating its glycoprotein nature. These results suggest that the circulating GM-CSF would contribute the increase of granulocyte-macrophage hemopoiesis in the early phase of malaria. (Summary) A neutrophil chemotactic factor (TfNCF) was isolated from the crude extract of Tritrichomonas foetus organisms by a combination of anion-exchange chromatography on DE52 and gel filtration on Sephacryl S200. TfNCF showed homogenicity by both PAGE and SDS-PAGE. The molecular weight of TfNCF was estimated to be 22 and 24 kDa, by Sephacryl S200 gel chromatography and by SDS-PAGE under reducing conditions. Immunization of TfNCF caused almost complete protection against T. foetus infection in mice. Western blot analysis probed with anti-TfNCF antibody showed that the epitopes on TfNCF were not commonly shared on the other components of T. foetus organisms nor other helminthous parasite-derived components. Furthermore, pre-incubation of neutrophils with antigens of other helminthous parasites or N-formylmethionyl-leucyl-phenylalanine did not affect the neutrophil chemotactic activity for TfNCF. These results suggest that TfNCF is a novel NCF consisting of unique epitopes for both antigenicity and neutrophil chemotactic activity. The role of NCF in the initiation of the immune response is discussed. (Summary) The immune responsiveness to specific antigens or mitogens was examined in jirds after primary and secondary infections with Brugia pahangi. When spleen cells were obtained from secondarily infected jirds, their proliferative responses to mitogens such as Con A or LPS, or to specific antigens prepared from infective larvae or adult worms were significantly lower than those of spleen cells obtained from primarily infected jirds. The proliferative responses of the peripheral blood mononuclear cells obtained from animals undergoing primary and secondary infections also showed a similar tendency. The depressed proliferative responses of the secondary infected spleen cells to Con A or LPS was partially restored by removing adherent/phagocytic cells from the original cell populations. After deletion of the adherent cells, however, antigen-specific proliferative responses were not altered and remained at low level. These results suggest that at least two different mechanisms of depression, namely adherent cell-mediated antigen-nonspecific suppression and unresponsiveness of lymphocytes to filarial antigens, are induced in jirds in the secondary infection. (Summary) In seven patients with paragonimiasis westermani, parasite-specific IgE and IgG levels in sera and pleural effusion were determined by an enzyme-linked immunosorbent assay (ELISA). The sensitivity to adult excretory-secretory (E-S) antigen was compared with the sensitivity to whole worm extract antigen, and the former was more sensitive in both an IgE-ELISA and IgG-ELISA. Both parasite-specific IgE and IgG could be detected by ELISA at levels much higher than those in control subjects using E-S antigen. When specific IgE and IgG levels in sera and pleural effusion of individual patients were compared, the latter had higher values. The difference between levels of specific IgE in pleural effusion and serum did not correlate with that of specific IgG. These results indicate that specific IgE and IgG antibodies form locally, i.e., in the lung, and that pleural effusions from patients with paragonimiasis are more suitable than serum for immunodiagnosis. (Summary) We examined the effect of adherent cells from bone marrow or spleen of mice infected with Plasmodium berghei on dyserythropoiesis. Significant reduction in number of erythroid progenitors (erythroid colony-forming units: CFU-E and erythroid burst-forming units: BFU-E) was observed in bone marrow as early as 1 day after P. berghei infection. When adherent cells were removed from bone marrow or spleen cells of infected mice, the number of CFU-E and BFU-E was clearly increased. Furthermore, addition of adherent cells from infected mice to nonadherent cells from normal mice inhibited erythroid colony formation significantly in a dose-dependent manner. These results suggest that the adherent cells obtained from bone marrow or spleen of mice in the early stage of P. berghei-infection have a suppressive effect on erythropoiesis. (Summary) The susceptibility of haemopoietic stem cell deficient W/Wv mice to infection with Plasmodium berghei was examined. The mean survival time of W/Wv mice after the infection was shorter than that of the +/+ mice. Splenomegaly, a characteristic pathological change of the host after infection with malaria parasites was not observed in W/Wv mice. When haemopoietic activity of the infected mice was examined, a substantial increase in number of multipotent haemopoietic stem cells (CFU-S) and the committed stem cells for granulocytes and macrophages (CFU-GM) or for erythrocytes (CFU-E) was observed in the bone marrow and spleen of +/+ but not of W/Wv mice. CFU-S were not detected in W/Wv mice before or after infection. The number of CFU-GM and CFU-E in bone marrow and spleen of W/Wv mice decreased after infection. Bone marrow grafting from +/+ to W/Wv mice 8 weeks before infection prolonged the mean survival time of the mice and effectively restored the number of CFU-S in the spleen of W/Wv mice. These results indicate that multi-potent haemopoietic stem cells play an important role in the host's defence mechanisms against P. berghei-infection. (Summary) Extramedullary hemopoiesis recognized as hemopoietic foci increased in the liver of Schistosoma japonicum-infected mice in parallel with kinetic changes in periovular granuloma formation. At the peak of the response, about 65% of the hepatic hemopoietic foci were of eosinophil lineage. When S. japonicum-infected mice were irradiated, hepatic hemopoietic foci rapidly disappeared within 3 days, whereas inflammatory cells in the periovular granulomas slowly reduced in number. When the number of hemopoietic stem cells in the liver were examined by spleen-colony assay, kinetic changes in the number of hemopoietic stem cells in hepatic nonparenchymal cells paralleled those of hepatic hemopoietic foci. Hemopoietic stem cells were rare in the granuloma cells. These results indicate that in response to the increased demand for eosinophils and other inflammatory cells, the liver acts as an extramedullary hemopoietic organ in which inflammatory cells are generated from hemopoietic stem cells. (Summary) Mechanisms of eosinophil response induced by soluble egg antigen (SEA) in Schistosoma japonicum-infected guinea pig tissues were analyzed using two purified components of SEA: major allergenic components (JEAL) and a major eosinophil chemotactic component (ECF-SjE). Eosinophil response to the crude SEA of S. japonicum was detected at the injection site mainly at 2-8 h after the antigenic challenge; this response consisted of two different phases of eosinophil attraction. An early phase (2 h) was mediated by an immediate-type hypersensitivity reaction; JEAL played an important role in mediating this phase of eosinophil response. A late phase (8 h) was mediated by ECF-SjE. Therefore, this phase of eosinophil response was caused by direct eosinophilotactic activity of SEA. These two types of eosinophil response must be important processes in the formation of S. japonicum egg-associated eosinophilic granuloma. (Summary) Eosinophil chemotactic activity in sera from mice undergoing an acute stage of schistosomiasis japonica and mansoni was examined. Eosinophilotactic activity in the serum was dependent on the dose and time of infection. Eosinophilotactic activity in sera from S. japonicum-infected mice was higher than that from S. mansoni-infected mice when they were compared at the comparable dose and time of infection. After gel chromatography on Sephadex G-200, eosinophilotactic activity in sera from mice infected with 30 cercariae of S. japonicum for 5 weeks was detected in the high molecular weight component. On the other hand, when sera from mice infected with 30 cercariae of S. japonicum for 8 weeks was chromatographed through Sephadex G-200 columns, eosinophilotactic activity was segregated into high (greater than 455,000) and low (less than 13,000) molecular weight components. High molecular weight ECF in sera from mice infected with 30 cercariae of S. japonicum for 8 weeks had high affinity to Con A, and was stable to heating or pronase digestion, but was sensitive to periodate oxidation, indicating its polysaccharide or glycoprotein nature. This high molecular weight ECF could be adsorbed by, and eluted from immunoaffinity beads coated with rabbit IgG anti-S. japonicum adult worm antibody. Thus, at least some part of circulating high molecular weight ECF would be derived from adult parasites. (Summary) High neutrophil and eosinophil chemotactic activities of soluble extract of Schistosoma japonicum cercariae (SjCe-ext) were detected in vitro by using blind-well chemotaxis chambers and Millipore filters in a dose-dependent manner. Low doses (total of 0.25 and 2.5 micrograms) of SjCe-ext induced significant neutrophilic infiltrations in normal and S. japonicum-infected (8 week) guinea pig tissues beginning at 2 hr after the intradermal injection. Eosinophilic responses were observed in the infected animals especially in early phase (2-8 hr post-injection of SjCe-ext). SjCe-ext elicited strong immediate skin reactions (at 30 min) in the infected guinea pigs. Possible roles of these chemotactic activities and hypersensitivity reaction in S. japonicum infection are discussed. (Summary) In order to determine whether the plerocercoid of Spirometra erinacei itself has eosinophil and neutrophil chemotactic factors, in vivo and in vitro examinations were carried out. We could observe large numbers of eosinophils and neutrophils which accumulated at the injection site of normal guinea pig skin following intradermal injection of soluble extract of plerocercoids of S. erinacei. At 1 hour after the injection, neutrophils appeared at the site, and the cell number reached its peak at 4 hours. Eosinophils appeared rather later than neutrophils (at 2-4 hours), and the number of cells reached its peak at 8 hours after the injection. Eosinophil and neutrophil chemotactic activities were also confirmed in an in vitro system by using a blind-well chemotaxis chamber with a Millipore filter in dose dependent fashion. An eosinophil chemotactic factor (ECF) with molecular weight of approximately 15,000, and two different neutrophil chemotactic factors, one of about same molecular weight as the ECF and the other of low molecular weight, were demonstrated by gel filtration on Sephadex G-200. Furthermore, it was confirmed that those factors were released from parasite by the detection of intensive eosinophil and neutrophil chemotactic activities in the culture supernatants containing plerocercoids. (Summary) Significant chemotactic activity for eosinophils was detected in soluble egg antigen (SEA) preparations of both Schistosoma japonicum and S. mansoni in dose-dependent fashion. The activity of S. japonicum SEA was higher than that of S. mansoni SEA. Gel filtration on Sephadex G-150 showed that S. japonicum SEA was composed of two groups of eosinophil chemotactic factors (ECFs), one of high molecular weight (JEE-H) and the other of low molecular weight (JEE-L). S. mansoni SEA showed ECF composition similar to that of S. japonicum SEA. JEE-H was stable on heating (100 degrees C, 60 min) and resistant to pronase digestion, but was sensitive to periodate oxidation. JEE-L was also stable on heating and resistant to pronase and carboxypeptidase A digestions. These properties of the ECFs were also held in common with those of S. mansoni SEA. JEE-L was extractable by toluene, indicating a hydrophobic nature. These results suggest that schistosome eggs themselves contain ECFs, and that the composition of S. mansoni and S. japonicum SEA-derived ECFs is essentially the same. However, they differ from the other ECFs which have already been described in schistosome infections. 1.	Makoto Ohashi, H Arita and A Niwa : Production of eosinophil chemotactic factor by CD8+ T-cells in Toxocara canis-infected mice., Parasitology Research, Vol.84, No.2, 136-138, 1998. (Summary) Production of eosinophil chemotactic factor by T-lymphocytes (ECF-L) was examined in Toxocara canis-infected mice. When spleen cells from T. canis-infected mice were cultured in serum-free RPMI1640, ECF-L production was detectable in an antigen-specific manner. The ECF-L production peaked at day 9 post-infection and then decreased. Depletion of Thy 1.2+ cells or CD8+ cells completely abrogated ECF-L production, whereas depletion of CD4+ cells did not, indicating that CD8+ T-cells are involved in the production of ECF-L. When bone marrow eosinophils obtained from T. canis-infected mice were preincubated with ECF-L, their chemotactic reactivity to parasite-derived ECFs was enhanced, whereas that of peritoneal cavity-derived eosinophils was not. Thus, ECF-L seems to be important not only as a chemoattractant but also as an activator of the chemotactic reactivity of naive eosinophils to the parasite-derived ECF in T. canis infection. (Summary) 「ものづくり」には，創造力育成の教育効果が期待されるが，これまで教養教育のカリキュラムの中で取り入れられることは，比較的稀であった．昨年度より徳島大学全学共通教育の創成学習の科目として，ものづくりを取り入れている．現代の社会的な背景とジェンダーの視点から「ものづくり」の意義を考えながら，教養教育における「ものづくり」の課題について検証する． (Keyword) general education / gender / ものづくり

(Summary) 平成17年度より，徳島大学全学共通教育の教養科目群にて従来の講義形式，ゼミナール形式に加えて，創成学習形式が開始された．この報告ではこれまで2年間行った『創成学習「つたえること」と「ものづくり」』について報告する．課題の提示方法を変えるなどの工夫を行ってきたが，学生の達成度を評価とともに，アンケートの集計結果から，今後の展望について考察してみたい． (Keyword) general education / innovation and creativity / communication
